Abstract
Objective:To explore the mechanism of QDTMP repairing function of apoptotic human umbilical vein endothelial cells(HUVEC)induced by hyperhomocysteine(HCY).Methods:Using the whole marrow-adherence way to get the bone marrow mesenchymal stem cell(MSCs)and collagen-digest way to get the HUVEC.FⅧ factor was stained to identify HUVEC.HUVEC were inoculated into the lower chamber of Taswell and intervene by HCY,24 h later MSCs were inoculated into the upper chamber.The percentage of HUVEC' apoptosis was investigated by flow cytometry;the conformation of apoptotic HUVEC was observed by electricity microscope.The expression of caspase-3、c-IAP-1、c-IAP-2 of HUVEC was investigated by west-blotting.Results:The apoptotic percentages of HUVEC in damaged(EC+HCY)group increased than that of normal control group(P0.01)and the apoptotic percentages of all the QDTMP intervene groups were lower than those of HUVEC+HCY group(P0.05~0.01).The expression of caspase-3 whose activity was restrained of QDTMP intervene groups decreased and the expression of their c-IAP-1 and c-IAP-2 increased compared with HUVEC+HCY group.In the entire group,the most obvious change was in 15% QDTMP group.Conclusion:Chinese prescription QDTMP can increases the expression of c-IAP-1 and c-IAP-2 to inhibit the activation of caspase-3,thereby inhibit the apoptosis processes and then protect the HUVEC.
