The Effect of Diet on Expression of Protein Tyrosine Phosphatase 1B in Liver of Insulin Resistant Rats

Abstract

Objective To investigate the effect of diet on the expression of protein tyrosine phosphatase (PTP) 1B in liver of insulin resistant rats induced by high fat diet. Methods 30 male Wistar rats were randomly divided into the control group (n=10) which was given basic diet, and the model group (n=20) which was given high fat diet. After 4 weeks, the model group was randomly divided into 2 subgroups:insulin resistant group which continued to receive high fat diet, and diet-treated group which accepted diet intervention for 6 weeks. The protein expression level of PTP1B in the liver of rats were determined with western blotting at the end of 10th week. Results The protein content of PTP1B in the liver of insulin resistant rats significantly increased by 98.3% compared with the control group (t=9.335,P0.001). After 6-week diet treatment, the protein level of PTP1B in the liver of diet-treated group obviously decreased by 32.7% compared with insulin resistant group(t=4.076,P0.001). Conclusion Diet treatment could improve insulin resistance possibly by reducing the protein expression of PTP 1B in the liver.
