Abstract

Objective To construct CD24 small interfering RNA (siRNA) expression vector and evaluate the inhibitory effect of siRNA on the expression of CD24 in cervical cancer cells line Hela.Methods The hairpin structure of siRNA tran-script template targeting CD24 gene was synthesized and inserted into pSilencer3.1-H1-Hypro plasmid, an eukaryotic expression vector.The recombinant plasmid was verified and transfect into Hela cells using Lipofectamine 2000.Expression of CD24 mRNA and protein were detected by RT-PCR and Western blot respectively.Results The designed nucleotide sequence was successfully inserted into the vector.Expression of CD24 was obviously down-regulated by pSi-CD24 compared with the control group at both mRNA and protein level.Conclusion CD24 siRNA eukaryotic expression vector was constructed successfully.The pSi-CD24 plasmid can inhibit the CD24 expression in Hela cells efficiently.
