Effect of MMP-1gene silencing by RNA interference on the migration and invasion of tongue squamous carcinoma cells

Abstract

Objective To investigate the effect of matrix metalloproteinase-1(MMP-1)-short hairpin RNA(shRNA)on the MMP-1expression,migration and invasion of human tongue squamous carcinoma cell line CAL27 in vitro.Methods shRNA was designed and synthesized to interfere MMP-1expression,which was ligated into PLKO.1vector for constructing recombinant lentiviral expression vector PLKO.1/MMP-1-shRNA.Then CAL27 cells were stably transfected with MMP-1-shRNA and the protein expression of MMP-1was detected by Western blot.The migration and invasion of CAL27 cells were determined by wound healing and Transwell assay,respectively.Results The recombinant lentiviral vector was successfully constructed,which could significantly downregulate MMP-1expression,inhibit the migration and invasion of CAL27 cells.Conclusions PLKO.1/MMP-1-shRNA vector can effectively inhibit MMP-1expression,cell migration and invasion.MMP-1 may be an important regulator in the metastasis of tongue squamous carcinoma cells.
