Model of rat's liver cirrhosis and portal hypertension induced by individuation of thioacetamide dosage

Abstract

Objective To develop an ideal means to induce rat's liver cirrhosis and portal hypertension by individuation of Thioacetamide(TAA) dosage based on rat's variations in response to TAA.Methods Fifty male SD rats were divided into three groups.Group 1(n=20) was treated with traditional means.Group 2(n=20) was treated with the dose modified according to weight changes during the induction of cirrhosis.Group 3(n=10) was control group.Results The mortality in group 1 was 30%(6/20) and cirrhosis developed was 50%(10/20).In contrast,the mortality in group 2 was 10%(2/20) and cirrhosis developed was 80%(16/20).The control group was all survival.The PVP and SPP of group 2 were higher than that of group 3 and MAP was lower than that of group 3(P0.05).STB and ALT were higher than that of group 3,but SAB,RBC,HB and PLT decreased obviously(P0.05).Conclusion Adjusting the inducible of TAA dosage based on rats' variations in response to TAA can successfully induce rat's liver cirrhosis and portal hypertension.Compared with the traditional method,our method can obviously reduce mortality and strikingly increase the production and quality of cirrhosis induced in the rat.
