Abstract
To investigate the possible preventive mechanism of Qichoutang on renal tubular epithelial cell transdifferentiation in renal interstitial fibrosis in rats.Using unilateral ureteral obstruction UUO induction renal interstitial fibrosis animal models with enalaprilat as the positive control medicine,rats were randomly divided into Sham,UUO,ACEI,Qichoutang of high-dosage,mid-dosage and low-dosage groups.Rats were killed in 12th day after surgery,Scr,BUN,CHO,TG,RBC and Hb were collected from each group.The expression of the renal interstitial α-smooth muscle actin α-SMA and Ⅲ collagen Col Ⅲ of rats from each group were detected by semi-quantitative immunohistochemical analysis.Measured the tubulointerstitial damage index and collagen deposition level of kidney tissues with HE,VG staining.In UUO rats,the tubular interstitial damage index,collagen deposition level,the expressions of α-SMA,collagenⅢ(Col Ⅲ),BUN,Scr,CHO,TG,RBC,Hb were all increased compared with thse of Sham group(P0.01).Qichoutang significantly decreases the expressions of α-SMA,ColⅢ,BUN,Scr,CHO,TG,RBC,Hb compared with ACEI rats(P0.05) and ameliorates tubulo interstitial damage index,collagen deposition.In renal interstitial fibrosis-induced UUO rats,Qichoutang may retard the progression of renal fibrosis by down regulation the expression of α-SMA,LN,ColⅢ,BUN,Scr,CHO,TG,decrease tubulointerstitial damage index and collagen deposition and prevent transdifferentiation action from renal tubular epithelial cells to mesenchyme cell.
