Abstract
Enzyme characteristics of protease from alkaliphile S9 were investigatedin this study. The protease was purified by means of ammonium sulfate precipitation and dialysis. Results showed that the optimal temperature and pH for hydrolysis of casein were 60 ℃ and 9.0 respectively. And the protease was stable at 40 ℃ and pH 8.0~10.0. Different metal ions showed different effects on the protease activity. K+, Ca2+, Mn2+ and Mg2+ could increase the protease activity, whereas Zn2+, Cu2+, Pb2+, Fe3+ and Hg2+ could cause inhibition.
