Abstract
In this report,four genotypes of maize(Zea mays L.)embryonic calli were transformed via Agrobacterium tumefaciens.The expression level of β-glucuronidase(Gus) gene was used as the evaluation index of influencing transformation efficiency.The results showed that there was dominant difference among different maize genotypes.The hybrid line Q31×Z31、Q31×Z3 and line B73 were more sensitive than line Z3.Improved transformation efficiencies were obtained when the embryonic calli were incubated with Agrobacterium suspension cells(OD600=0.4~0.6) for 10 min in the infection medium.Optimized co-culture condition was that infection was performed at 23℃ in dark for 3 days.The transgenic maize plants with basta-resistant were obtained via Agrobacterium-mediated transformation.
