Abstract
OBJECTIVE:To investigate the inhibition effect of αvβ6 siRNA on invasiveness of human ovarian cancer cells HO-8910PM.METHODS:With liposome as a vecter,integrin alphav beta6 siRNA and non-oligonucleotide were transfected into HO-8910PM cells,which were then randomized to siRNA group,Neo group and control group.The expressions of integrin αvβ6 mRNA and protein were determined by quantitive RT-PCR and Western-Blot,and the proliferation and invasion were measured by MTT and Transwell ventricle.Then cells were inoculated subcutaneously into the left flank of nude mice.The growth rate was measured and expressions of integrin αvβ6 in transplanted tumors of nude mice were detected by immunohistochemical method.RESULTS:Compared with the Neo group and blank control,the expressions of integrin αvβ6 mRNA and protein in the siRNA group were obviously lower;the invasiveness of human ovarian cancer cells transfected by integrin αvβ6 siRNA was obviously decreased (P0.01).The proliferative activity of HO-8910PM cells in vitro was not affected by transfection with the alphav beta6 siRNA (P0.05).Inhibition of αvβ6 expression in HO-8910PM reduced the cell invasiveness.Furthermore,cells transfected with αvβ6 siRNA formed smaller tumor in transplanted tumors of nude mice than Neo and control group.CONCLUSION:Integrin alphav beta6 siRNA can inhibit invasiveness of human ovarian cancer cells,and may be of great benefit for finding rational approach in ovarian cancer therapy.
