Abstract
Objective To observe the effects of 3-n-butylphthalid(NBP) on the behaviors and the expression of ICAM-1 in hippocampus of vascular dementia(VD) model.Methods Kunming mice were divided randomly into three groups:model group,sham-operated group and treated group(NBP,7.5 mg·kg-1·d-1).The mice model was established by repeated occlusion and reperfusion of the bilateral common carotid arteries and ischemic hypotension for three times.The capabilities of learning and memory of mice were investigated by the step down test and water maze test,the change of ICAM-1 exprssion in hippocampus CA1 in each group was detected by immunohistochemistry technique.Results Compared with sham-operated group,the abilities to learning and memory in model group decreased markedly(P0.05),and the number of immunoreactive positive neurons of ICAM-1 increased significantly(P0.01).The grades of learning and memory in NBP-treated group ameliorated apparently compared with model group(P0.05),and the number of immunoreactive positive neurons of ICAM-1 decreased meanwhile(P0.01).Conclusion NBP could inhibit the inflammatory response in VD by reducing the expression of ICAM-1 in hippocampus,thereby reduced the extent of brain damage and improved the clinical symptoms of VD.
