Effect of Ulinastatin on heat shock protein 70 expression in rats with septic acute lung injury

Abstract

Objective To study heat shock protein 70(HSP70) expression and investigate the possible mechanisms of Ulinastatin therapy in rats with septic acute lung injury(ALI).Methods Thirty-nine Wistar rats were randomly divided into sham group(n = 12),ALI group(n = 15) and Ulinastatin(UTI) group(n = 15).And rats were sacrificed at 6,12 and 24 hours after cecal ligation and puncture operation.Arterial blood gas analysis,lung tissue wet/dry weight ratio,lung histopathologica1 changes and pulmonary expression of HSP70 mRNA assessed by realtime-PCR were observed.Results Compared with ALI group,PaO2 in UTI group was significantly increased whereas the ratio of wet/dry weight was decreased(P 0.05).The grading of pathologic damage of lung tissue was attenuated greatly.Pulmonary expression of HSP70 mRNA were increased in UTI group compared to those in ALI group at all time points(P 0.05).Conclusion Effects of Ulinastatinon on the improvement of septic acute lung injury may be attributed to the up-regulation of lung HSP70 expressions.
