Abstract
Objective To investigate the effects on neuronal cell apoptosis of frontal lobe and P53 protein expression after global ischemia and reperfusion.Methods The acute global ischemia(15 min) model was produced by four- vessel occlusion as described by Pulsinelli in Sprague -Daewley rats, after cerebral ischemia, the rats randomly divided into three groups, that is normal group( n = 7 ) , sham - operative group( n = 49) , operative group(n= 49),all animals were decapitated in 1 、6、12、24、48、72h and 7d,respectively,after reperfusion.The neuronal cell apoptosis were detected by TUNEL method,meanwhile we utilized immunohitochemistry to observe the expression of r protein in frontal lobe. Results There were a few TUNEL - positive cells in 24h after ischemia and reperfusion, we observed more TUNEL - positive cells in 48h, this expression was maximal in 72h,7d after ischemia TUNEL- positive cells declined obviously.Immunohitochemistry staining:there were a few P53 protein expression in 24h in ischemia group.This expression was maximal in 48h, subsequently began to decline,a few of expression were observed at 7d after ischemia,The staining mainly found in the nuclei.Conclusion The delayed neuronal death after global ischemia and reperfusion was apoptosis,P53 protein expression was increased following global ischemia and reperfusion, the relationship between the neuronal cell apoptosis and P53 protein expression was a positive correlation in a certain time following global ischemia and reperfusion.
