Abstract
Objective:To investigate the relationship between HPV 16 infection and the expression of P21~(waf) and Ki67 in cervical intraepithelial neoplasia(CIN) and squamous cell carcinomas(SCC) of the cervix uteri.Method:Tissue microarray combined with in situhybridization(ISH) and immunohistochemical staining(EliVison)~(tx) plus method was used to detect the expression of HPV16 RNA,Ki67 and P21~(waf) protein in all 130 cases of normal cervical tissue and CIN and SCC of the cervix uteri.Results:1)The positive rate of HPV16 hibrid signals in CINⅡ,CINⅢ,In situ carcinoma and SCC were all significantly higher than in normal cervical tissue(all P0.05),and was also higher in SCC than in CIN(χ~2=5.670,P=0.017).2)The positive expression rate of P21~(waf) protein in SCC were significantly lower than normal cervical tissue(P0.05),but no significant difference between in SCC and in CIN(P0.05).It was also found a negtive correlation between expression of P21~(waf) and infection of HPV16(P0.05,r=-0.337).3)There was a significant difference in the expression of Ki67 between normal cervical tissue and CINⅡ,CINⅢ,In situ cancer,SCC respectively(all P0.05).4)The positive expression of P21~(waf) and Ki67 showed negative correlation in the progress form CIN to SCC(P0.05,r=-0.446).Conclusion:There are very important relationship between HPV16 infection and the negtive or poor expression of P21~(waf) and the high expression of Ki67 in the progress from CIN to SCC.HPV16,Ki67 show negative correlation to P21~(waf).Maybe it can reveal the detailed mechanism which HPV 16 participate in malignant transformation and give some reference to improve the early diagnosis of cervical cancer.Tissue microarray is an efficient technique to study genes and their expression product.
