Abstract
Using transcription of T7RNA polymerase in vitro,two complementary single stranded RNAs(ssRNAs) directing to the template RNA of telomerase(hTR) were synthesized and then annealed to form the small interfering RNA(siRNA).Telomeric repeat amplification protocol(TRAP) was applied to detect the telomerase activity and analyze the interfering effects of siRNA in lysate of tumer cells.The results indicated that T7 RNA polymerase can efficiently transcribe short ssRNAs.The siRNA prepared could notably decrease the telomerase activity in tumor cells.This decrease was much stronger than that displayed by the equal amount of antisense RNA strand.The technique of siRNA synthesis with T7RNA polymerase is a cheap,effective,simple and high-output way.It could provide a new exploring approach for the gene therapy of neoplasms.
