Abstract

Objective To investigate the expression of myocardial sarcoplasmic reticulum calcium handling correlative proteins(sarcoplasmic reticulum calcium adenosine triphosphatase,SERCA2,and phospholamban,PLB)gene in heart failure rabbit and the effects of valsartan on the prevention of chronic heart failure.Methods Eighteen rabbits were divided into three groups:6 rabbits with heart failure induced by volume plus pressure overload,6 sham operated rabbits and 6 rabbits treated with valsartan.Seven weeks later,left ventricular function,hemodynamic parameters,and SERCA2,PLB gene expression were observed.Results Compared with the sham operated rabbits,HR,LVMI and LVEDP in heart failure rabbits were significantly increased(P0.05),while their left ventricular shorten fraction and ejection fraction were decreased(P0.05).Compared with the heart failure rabbits,HR,LVMI and LVEDP in the valsartan treated rabbits were significantly decreased(P0.05),while their left ventricular shorten fraction and ejection fraction were increased(P0.05).SERCA2 and PLB gene expression in heart failure rabbits were remarkably lower than that in sham operated rabbits and valsartan treated rabbits(P0.05).Conclusion Valsartan can improve cardiac function,probably owing to its upregulating gene expression of myocardial sarcoplasmic reticulum calcium handling correlative proteins(SERCA2 and PLB).
