Tetraploid induction and identification of Anthurium scherzerinum

Abstract

Calli of two diploid varieties of ‘Smaragd’ and ‘Artus’,were employed to the tetraploid induction of Anthurium scherzerinum by colchicine treatment method.The results showed that the induction rate depengded on the genotype of A.scherzerinum,the concentrations of colchicene treatment and duration of treatment.The induction rate was different between different genotypes.The lower the colchicine concentration was,the higher the induction rate was.At the same concentration,the longer the duration of treatment was,the higher the induction rate was.The highest induction rate of the two genotypes was 52.5% and 61.90%,separately,which were cultured on the liquid medium contained 0.2 g/L colchicine for 14 d on a rotator.The higher the concentration of colchicene treatment was,the lower the absolute growth rate and the average amout of bud differenation were.The improved squash method was the accurately method of the tetraploid identification of A.scherzerinum.
