Effects of nm23-H1 gene transfection on in vitro invasion potential of human cholangiocarcinoma cell line QBC_(939)

Abstract

Objective To explore the role of nm23 H1 gene overexpression in invasion potential of human cholangiocarcinoma cell line for its potential application in gene therapy for cancer. Methods A mammalian expression vector containing nm23 H1 cDNA was transfected into human cholangiocarcinoma cell line QBC 939 . Northern blotting and immunohistochemistry were used to analyze the expression levels of nm23 H1 and MMP 9, and in vitro invasion potentials the tumor cells before and after the gene transfection. Results After transfection, the nm23 H1 mRNA and protein expression was up regulated significantly. The ability of in vitro invasion through matrigel was remarkably decreased. Immunohistochemical staining showed the expression of MMP 9 were decreased after transfection. Conclusion Up regulation of nm23 H1 in cholangiocarcinoma cell line suppresses its ability of in vitro invasion and reduces the expression of MMP 9 in it.
