Effects of c-myc antisense oligonucleotide on the proliferation and apoptosis of osteosarcoma cells

Abstract

Objective To study the effects of c-myc antisense oligonucleotide on the proliferation and apoptosis of osteosarcoma cells(MG-63). Methods Designed c-myc antisense oligonucleotide fragment was transfected into human osteosarcoma MG-63 cell and its effects on the proliferation、apoptosis were observed by MTT method and HE stain and transmission electron microscope. Results It showed that c-myc antisense oligonucleotide was able to inhibit the proliferation of human osteosarcoma MG-63 cell at the doees of 10.0μmol/L and effected for forty-eight hours; Cells exhibited characteristic morphologic changes of apoptosis. FCM method checked human osteosarcoma MG-63 cell into the S phase induce apoptosis (the apoptosis rate could be to 36.82%) and inhibit the expression of c-myc gene protein. Conclusion c-myc antisense oligonucleotide is able to induce apoptosis of human osteosarcoma MG-63 cell and inhibit the proliferation of human osteosarcoma MG-63 cell.
