Abstract

OBJECTIVE To evaluate the effect of valsartan a specific type angioteinsin II receptor (AT 1 ) antagonist, on lectin like oxidized low density lipoprotein receptor 1 (LOX 1) mRNA and protein expression in cultured THP 1 cells METHODS Valsartan was added to the culture medium for 30 min before cells were incubated with Ang II(1 0×10 -6 mol·L -1 )for 24 h LOX 1 protein expression was detected by cell enzyme linked immunosorbent assay (ELISA), and LOX 1 mRNA expression was detected by quantitative competitive reverse transcription polymerase chain reaction (RT PCR) RESULTS 1 0×10 -5 mol·L -1 valsartan blocked Ang II induced LOX 1 protein and mRNA expression in THP 1 cells completely 1 0×10 -6 ,1 0×10 -7 ,1 0×10 -8 and 1 0×10 -9 mol·L -1 valsartan decreased Ang II induced LOX 1 protein expression by 100%, 85 8%, 47 2% and 9 8% respectively LOX 1 mRNA expression was blocked by valsartan for 95 1%, 82 6%, 63 0% and 20 0% in each concentration CONCLUSION Valsartan blocked Ang II induced LOX 1 protein and mRNA expression in THP 1 cells This may be related to the mechanisms of anti atherosclerotic properties of valsartan
