Abstract

Artificial basement membrane (ABM) with spongiform configuration was mainly made of type I collagen (rat tail) which was formed into hydrated collagen lattices (HCLS), fibronectin, type IV collagen and laminin. ABM could cut off 10% of the fetal bovine serum (FBS) in medium, raise the viability of cells and prolong the period of subculture. The growth effecien-cy, plating effeciency and the rate of clonal growth of CNE-2 cells propagated on ABM with 2-5% FBS cultural medium were higher but the uptake of 3H-TdR was lower than that of these cells on glass dishes with the same cultural medium. These findings suggested that the lower concentration FBS of medium should be used for researching the effect of extracellular matrix on cells, and that the ABM is idealer matrix of culture for inducing the differentiation of carcinoma cells in vitro.
