Abstract

Objective To evaluate the effect of controlled-rate freezing in autologous mobilized peripheral blood stem cells(PBSC). Methods Mobilized PBSC were cryopreserved in liquid nitrogen with controlled-rate freezing to -80 ℃. Patients were infused their autologous PBSC after myeloablative conditioning regimen. Nucleated cells and CFU-GM was tested before and after thaw. Excluded trypan blue was also tested after thaw. Results The average concentration of nucleated cell was 1.405 × 108ml with cryopreseration. The mean postthaw recoveries of nucleated cell and CFU-GM were 80.64 % ± 11.51 % and 94.01 % ± 25.42 %. Trypan blue dye exclusion was 84.69 % ± 8.72 %. All of the 26 patients established hematopoietic and immune reconstitution rapidly and stably except one dying of the toxicity of conditioning. Conclusion Controlled-rate freezing is a safe and effective cryopreservation technique for high concentration PBSC.
