Abstract

Purpose To screen cervical cancer-related microRNAs(miRNA) and predict selected miRNA targets.Methods Gene chip technology was used to detect expression of 1 891 of miRNAs in cervical cancer tissues and their adjacent tissues.Reliability of miRNAs microarray results was verified by RT-PCR.Software was applied to predict target genes of significantly changed miRNAs.Results Compared with adjacent cervical cancer tissues,27 differentally expressing miRNAs were obtained by screening,of which 3(hsa-miR-891a,hsa-miR-937,hsa-miR-F114) were upregulated by more than 1.5-fold,24(including hsa-miR-126,hsa-miR125a5p,hsa-miR-26a,hsa-let-7c) were downregulated by more than 1.5 times.RT-PCR results confirmed that hsa-miR-126 and hsa-miR26a were significantly reduced,consistenting with gene microarray results.But hsa-miR125a-5p expression was not significantly reduced.miRNA target prediction analysis demonstrated that miR-126 has hundreds of target genes.Conclusion The miRNA differential expression profiles of human cervical cancer may be obtained by gene microarray and RT-PCR,and suggests that hsa-miR-126 and hsa-miR-26a may be related with the occurrence and development of cervical cancer.
