Molecular Mechanism of Protective Effect of Total Saponins of Panax Notoginseny against Myocardial Ischemia Reperfusion Injury

Abstract

Objective:To study the changes of activation of nuclear factor κB(NF κB) ,expression of intercellular adhesion molecule 1(ICAM 1) ,infiltration of polymorphonuclear neutrophils(PMNs) and protective effect of total saponins of panax notoginseny (PNS)against myocardial ischemia reperfusion injury.Method:152 Newzeland rabbits were divided into three groups: Ischemia reperfusion group(IR group):Rabbits were subjected to total occlusion 45 min of left anterior desending artery(LAD) followed reperfusion; PNS preconditioning group(IR+PNS group): PNS 100 mg/kg pretreated rabbits 10 min before ischemia; and sham group.The myocardial samples were studied before ischemia (0),30,60,90,120,240,360 min after ischemia reperfusion.The expression levels of ICAM 1 were evaluated by Western blot,activation of NF κB by electrophoretic mobility shift assay(EMSA),the infiltration of PMN were measured by assaying the activity of myeloperoxidase.Result: In IR group,after ischemia reperfusion 30 min,activation of NF κB were increased significantly and reached a peak at 120 min,then decreased;the expression levels of ICAM 1 of myocardium were increased at 120 min and had positive correlation to the infiltration of PMN;In IR+PNS group,PNS could inhibit the activation of NF κB,expression of ICAM 1 and infiltration of PMN. Conclusion: The inducible activation of NF κB starts up ICAM 1 expression of myocardium and it is an important factor in myocardial ischemia reperfusion injury, PNS has protective effect on myocardium by inhibiting activation of NF κB ,ICAM 1 expression,and infiltration of PMN.
