Abstract
[Objective] It had the important significance in the improvement of vaccine′s protection efficiency to develop the preparation method of canine distemper virus(CDV) with high titer.[Method] The FK81 cells were cultured in the medium-MEM and mixed medium(MEM and DMEM was mixed by 9∶1) and inoculated with CDV and then the number of total cell, the amount of half cell infected by CDV(TCID50) and the morphology of virus particle under electron microscopy in 2 media were compared.[Result]The number of total cell,TCID50 and the morphology of virus particle in mixed medium were superior in MEM medium. It was also found in the determination of TCID50 of CDV,the method of enzyme-linked immunosorbent assay(ELISA) was more sensitive than cytopathic(CPE) method.[Conclusion] The method of CDV proliferation, established in the experiment, could be used in practice.
