Abstract
[Purpose] To study the effect of self-assembly multiplayer microcapsules loading doxorubicin on rabbit VX2 transplanted hepatic tumor model. [Methods] Eighteen New Zealand white rabbits im-planted VX2 carcinoma tissue into the left lobe of the liver, and they were divided into three groups. Each group consist of six rabbits. Ten days later the diameter of the tumor reach to 1.0 centimeter, the rabbits were given different drugs by intra-arterial hepatic administration(IAH).Group A were given natural saline; Group B doxorubicin(4mg/kg); Group C, microcapsules loading doxorubicin(equivalent dose 4mg/kg). PET/ CT was used to monitoring the treatment response of the tumor at 1-,3-,5-,7-,10-,14-day after treatment. The volume of the tumor, the tumor growth rate and the mean maximum FDG uptake (standardized uptake value,SUVmax) were measured. Animals were sacrificed 14 days after treatment, all groups were histologi-cally examined. [Results] The tumor volume and the growth rate (TGR) at 1 week after treatment in group B and group C were both significantly lower than those in group A (P0.05). Ten days after treatment, the tumor volume and tumor growth rate and the level of SUVmax was lowest in group C compared with the other groups(P0.05). Apoptosis cell increased in HE staining slice in group C. [Conclusion] Self-as-sembly multiplayer microcapsules loading doxorubicin can inhibit the cellular glycometabolism and sup-press the growth of the rabbit VX2 heptic tumor, induce the apoptosis of the tumor, resulted in increasing efficacy in chemotherapy for liver cancer.
