Abstract

Objective To obtain stable labelling of mesenchymal stem cells(MSCs) mediated by recombinant replication-deficient retroviral vector expressing green fluorescent protein(Rt-GFP) and provide a basis for in vivo differentiation study of MSCs in future.Methods Rat MSCs were cultured and purified with whole bone marrow method.The purified MSCs were infected with Rt-GFP.The GFP positive MSCs were observed with fluorescent microscopy.Results After Rt-GFP infection,80%~90% MSCs were labelled by GFP.The fluorescent intensity of labelled MSCs was not changeable with the prolonged culture time and the increase of passage.Conclusion MSCs may be stably labelled by Rt-GFP.
