Abstract
The purpose of this study was to investigate the scavenging activities(SA) of hydrolysates,which were produced by different hydrolytic enzymes from Cyrenodonax formosanus protein,on hydroxyl free radical by Fenton reaction,the active peptides produced by hydrolysis with Sephadex G-25 were separated,and the molecular weights distribution of the active peptides was measured.The results showed that the optimal enzymatic hydrolysis condition that could produce the protein hydrolysate with the highest scavenging activity on hydroxyl free radical for using papain was temperature 50℃,time 30 min,pH 7.5,enzyme content 0.15% and enzyme/substance ratio 1∶2(quality ration), and the scavenging activity(SA) was up to 86.9%;And for using trypsin were enzyme content 0.30%,time 85 min,pH 8.0,temperature 55 ℃ and substance concentration 1∶2(quality ration),and the scavenging activity(SA) reached 89.5%.The SA of fractionated active peptide,which was separated from the Cyrendonax formosanus protein hydrolysates produced by papain with the molecular weights 5.68×103,was 84.73%.And the Cyrenodonax formosanus protein hydrolysates produced by trypsin had two absorption bands,the fractionated component at the higher absorption band had the low scavenging activity on hydroxyl free radical,and at the lower absorption band with its molecular weights 1.165×104 had,SA was 88.49%,on hydroxyl free radical.
