Killing effect of anti-HER2 monoclonal antibody-TAX immunoconjugate on the tumor cells in vitro

Abstract

OBJECTIVE To study the specific killing effect of the anti-HER2 monoclonal antibody-TAX immunoconjugate on the tumor cells expressing HER2.METHODS Anti-HER2 monoclonal antibody(Sc7301) was conjugated to TAX via active ester method;The immunological activity of the antibody in conjugate was assessed by ELISA;The cytotoxicity in vitro of conjugate was determined by MTT method;the apoptosis rate of cells was observed using flow cytometer(FCM).RESULTS The immunological activity of antibody in the conjugate retained more than 54.8% of the original McAb Sc7301;The cytotoxicity in vitro of the conjugate to SKOV3 cell was nearly 2~3-fold more potent than the mixture,but the cytotoxicity of the conjugate to HepG2 was similar as the mixture of the TAX and Sc7301;flow cytometer analysis showed that the high the concentration of immunoconjugate,the more typical the subdiploid peak of the cells.CONCLUSION The immunoconjugate of anti-HER2 monoclonal antibody-TAX can specifically kill HER positive cells in vitro.
