Abstract

Objective To study optic nerve myelin staining methods so the best method for clinical diagnosis and mechanism research can be identified. Methods Three myelin staining methods were compared in this study for specificity and contrast: Weil hematoxylin, fast green and luxol fast blue. Results The Weil hematoxylin, fast green and luxol fast blue methods produced myelin stain colors of black, green and blue, respectively. Conclusion The Weil hematoxylin staining procedure was tedious, and the definition of fast green staining was not as good as the luxol fast blue. Therefore, the best optic nerve myelin staining method is luxol fast blue.
