Abstract

Objective: To investigate the effect of palmitate on accumulation of cytoplasmic triglyceride levels,and to establish the model of hypertrophic 3T3-L1 adipocyte.Methods: The 3T3-L1 adipocytes were treated with different concentration(0.0,0.1,0.2 and 0.4 mmol/L)of palmitate for 48 hours.The adipocytes were identified by red O staining.The levels of cytoplasmic triglyceride were determined by tissue triglyceride assay kit and adjusted by totally cytoplasmic protein concentration.Results: The levels of cytoplasmic triglyceride treated with different concentration(0.0,0.1,0.2 and 0.4 mmol/L)of palmitate were(5.0±0.6),(6.9±1.4),(5.7±0.5)and(5.6±0.7)μmol/mg respectively.Results: The 3T3-L1 adipocytes treated with of 0.1 mmol/L palmitate led to obviously accumulation of triglyceride in cytoplasm compared to the control.Conclusion: Palmitate with the concentration of 0.1 mmol/L leads to obviously accumulation of triglyceride in cytoplasm of the 3T3-L1 adipocyte.
