Abstract
Objective To study the effect of curcumin on apopsis and bcl-2 expression in SW480 human colon cancer cells. Methods The inhibitive effect of curcumin on SW480 was measured with MTT. Cell cycle phase was analysised by flow cytometry. Apoptosis of SW480 was studied by TUNEL. The expression of bcl-2 gene was determined by immunohistochemical analysis. Results MTT test showed that curcumin was cytotoxic to cells .the number of the apoptosis cell line increased.FCM showed that the proprortion of G_ 0/G_ 1 phase increased while the S and G_ 2/M phase decreased in the cell cycle. In curcumin treated cell line,bcl-2 expression was down regulated at protein level. Conclusions Curcumin inhibited the proliferation of SW480 by inducing apoptosis in the study. The prevented cells from entering the next cell cycle and down regulation of bcl-2 protein may be the mechanism.
