Abstract

To investigate the i nhibitory effect of Arsenic trioxide on GLC-82cells(lung cancer strain)and its m echanism.GLC-82cells in culture medium in vitro were given different c oncentrations of Arsenic trioxide.The inhibitory ratio of the cells were measure d by MTT assay,apoptosis was obsevered by flow cytometry(FCM)and hoechst33258flu orescence staining,and the activity of telomerase was detected using TRAP-PCR-ELISA before and after apoptosis occurred.Arsenic trioxide could decrea se the activity of telomerase,as well as cause apoptosis and inhibit the growth of GLC-82cells significantly.The suppression was both in time-and dose-depend ent manner.[Conclusion]Arsenic trioxide has anti-proliferation effect on lung c ancer GLC-82cells in vitro,inducing apoptosis and decreasing the activity of te lomerase.It may be one of the most important mechanisms.
