Abstract
Objective To investigate the occurrence of germ cell apoptosis and its mechanisms after lead administration in the tests of the adult male rats.Methods Adult male Wistar rats were treated with 5,10,20 mg·kg -1 body weight of lead for 3 weeks.The apoptosis of germ cell were assessed by employing TdT-mediated dUTP-biotin nick end labeling (TUNEL) technique.Immunohistochemical staining was used to detect Bcl-2 and Bax gene expression.Results In the tests of lead-treated rats,the apoptosis index of germ cell and Bax expression significantly increased compared with that in the control group(P0.01) while Bcl-2 expression obviously decreased(P0.01).Conclusion The spermatogenic cell apoptosis is highly associated with lead loading and Bcl-2/Bax may play an important role in it.
