Genetic Diversity and Genetic Relationship Analysis of 31 Alfalfa Accessions Based on RAPD Markers

Abstract

RAPD(Random amplification polymorphic DNA) molecular marker technique was used to detect the genetic diversity and phylogenetic relationships of 31 alfalfa accessions.341 individuals were sampled from 31 alfalfa accessions,and 11 individuals were sampled from each accession.123 bands were amplified with eight RAPD primers,121 bands were polymorphic and the percentage of polymorphic bands was 98.37%.Shannon information index was 0.4572.Nei's gene diversity index was 0.2967.The genetic differentiation coefficient among populations was 0.2697,the results indicated that alfalfa genetic variation mainly existed within accessions.All 31 accessions were clustered into 5 groups,genetic relationship between accession 25 and accession 42 was the nearest,genetic relationship between accession 2 and accession 17 was the widest.Accession27 was a special germplasm of M.sativa,accession 2 was a special germplasm of M.falcata.
