The effect of chrysin on in vitro activation,proliferation and cell-cycle distribution of the mouse T lymphocytes

Abstract

Aim: To investigate the effect of chrysin on the activation,proliferation and the cell cycle distribution of the mouse T lymphocytes in vitro and to elucidate its immunosuppressive mechanism.Methods: Fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the expression of CD69 by activated mouse T lymphocytes.the proliferation rate of mouse lymphocytes was analyzed by CFSE staining together with flow cytometry.The distribution of the cell cycle was analyzed by PI staining.Results: The expression rate of CD69 in activated T cells in response to ConA was(85.12±1.07)%.After treatment with 5 μmol/L、25 μmol/L、50 μmol/L CR,the expression rate of CD69 reduced obviously.The exprecsion rate was(78.42±1.82)%、(66.24±1.43)%和(53.09±1.77)% respectly.CR reduced the generations and the numbers of T lymphocytes induced by ConA.The cell-cycle distribution revealed that the cells number in G0/G1 was increased but in S was decreased.The cells number in G2/M was also decreased when CR was(25 μmol/L).Conclusion: CR can inhibit the activation and(prolife-)ration of the T lymphocytes and arreste activated T lymphocytes in G1. CR is a potential immunosupressive agents.
