Abstract
Objective To investigate the effect of hypoxia on HIF-1α expression in tongue squamous cancer cell line Tca8113 cells.Methods Tca8113 cells in logarithmic grow phase were cultured in hypoxia(1%O2、5%CO2、94%N2) conditions for 1/2 h,1 h,3 h,6 h,12 h and 24 h,respectively.Expression of HIF-1α mRNA were assessed by RT-PCR technique.Results The expression of HIF-1α mRNA increased significantly and reached the peack after 12 h of hypoxia,and then decreased to the level of normoxic conditions at 24 h.The expression of HIF-1α mRNA in hypoxia was higher than that in normoxic control groups.Conclusion Hypoxia up-regulates the expression of HIF-1α mRNA,which makes cancer cells to adopt hypoxia in solid tumor,and to promote tumor development.
