Expression of osteopontin in the lung tissue of acute lung injury rats and the influence of osteopontin on the expression of TNF-α and IL-10

Abstract

Objective:To investigate the expression of osteopontin(OPN) in the lung tissue of acute lung injury(ALI) rats induced by lipopolysaccharide(LPS) and explore the influence of OPN on the level of TNF-α and IL-10 in serum.Methods:56 male SD rats were randomly divided into 3 groups:control group(8 rats),ALI group(24 rats) and intervention group(24 rats) Then ALI group and intervention group were divided into 3 subgroups respectively and 8 rats in each subgroup were examined at 2,4 and 8 h.Rats were injected with 1 ml normal sodium(NS) in control group,with 1ml LPS solution(including LPS 6 mg/kg) in ALI group and intervention group via caudal vein.Five minutes later,rats in intervention group were injected with 0.5 ml anti-OPN antibody(1∶32) while rats in control group and ALI group were injected with 0.5 ml NS in the same way.PaO2 and Wet/Dry weight(W/D) were detected.Pathological changes of the lung tissue and ALI score were examined.The level of TNF-α and IL-10 in serum were detected by ELISA and the expression of OPN in homogenate of lung tissue were determined by Western blot at 2,4 and 8 h.Results:PaO2 was decreased in ALI group at 2,4 and 8 h compared with that of control group respectively(P0.01),but was increased in intervention group as compare to that of ALI group at the same time point(P0.05).W/D and ALI score in ALI group at 2,4 and 8 h were higher than those in control group(P0.01) and intervention group at the same time point(P0.05).The level of IL-10(pg/ml)in serum in ALI group at 2,4 and 8 h was remarkably higher than that in control group(P0.01),but was lower compared with intervention group at the same time point(P0.05).The level of TNF-α(pg/ml)in serum and TNF-α/IL-10 at 2,4 and 8 h in ALI group were higher than those in control group(P0.01) and intervention group at the same time point(P0.01).Compared with that of control group,the expression of OPN in homogenate of lung tissue in ALI group at 2,4 and 8 h was higher(P0.01),but there was no difference between ALI group and intervention group at the same time point(P0.05).Conclusion:The expression of OPN in lung tissue of ALI rats induced by LPS is increased.OPN can aggravate the pathology changes of ALI rats,which might be related to up-regulating expression of TNF-α and down-regulating expression of IL-10 by OPN.
