Abstract
Time-course changes of choline acetyltransferase (ChAT) activity and protein content in cultured frontal cortex neurons of fetal rats and the effects of nerve growth factor (NGF) on ChAT activity and protein content were studied. The activity of the ChAT was determined by spectrophotometric method, and protein content was assayed by Folin phenol method. With the prolongation of cultural time, ChAT activity and protein content increased gradually in the frontal cortex cultures. No significant effects of NGF (10 μg·L1 ) on ChAT activity were observed during the first week, but there was a marked increase of ChAT activity in NGF group after 7 d in culture. NGF (0.1-100 μg·L1) concentrationdependently increased the activity of ChAT and protein content in 13 d of plating. The results suggest that NGF accelerate central cholinergic neuron development and differentiation.
