Protective effects of low concentrations of rifampicin on rotenone-induced SH-SY5Y cells model of Parkinson′s disease

Abstract

Objective To explore the effects of low concentrations of rifampicin on human neuroblastoma(SH-SY5Y) cells.Methods The cells model of Parkinson′s disease was established by damaging SH-SY5Y cells with rotenone in vitro.Cell morphology was observed by microscope.Cell viability was measured using MTT assay.Intracellular reactive oxygen species(ROS) and apoptosis rate were measured by flow cytometry.α-Synuclein mRNA was measured with RT-PCR.ResultsCompared with rotenone injury control group,ROS,α-synuclein mRNA and apoptosis rate in the groups of pretreated with different concentrations(1,10,100 nmol/L) of rifampicin were significantly decreased(P0.01).Conclusion Rifampicin has the effects of anti-oxidative stress,reducing the α-synuclein mRNA level,and protecting SH-SY5Y cells from apoptosis.
