Abstract
Objective To explore whether or not the function of interferon γ(IFN γ) in promoting osteopontin gene expression and protein synthesis of rat vascular smooth muscle cell.Methods Cultured cells were incubated with different concentration of IFN γ for different hours, then RT PCR and Western blotting analysis were used to observe the effects of IFN γ on osteopontin gene expression of rat vascular smooth muscle cells Results IFN γ(1 000 U/ml )could obviously induce gene expression and protein synthesis of osteopontin, RT PCR results showed that the ratio of mRNA of osteopontin and β actin in cells incubated with IFN γ for 24 and 48 hours increased by 24 49%, 28 03% respectively, as compared with that of the control ( P 0 05), and Western blotting analysis also showed that protein synthesis of osteopontin in cells incubated with IFN γ for 24 and 48 hours increased by 71 18%, 75 66% respectively, as compared with that of the control ( P 0 05) Conclusion IFN γ could stimulate osteopontin gene expression and protein synthesis of rat vascular smooth muscle cells
