Abstract
Papain was immobilized on 0.5%(v/v)glutaraldehyde cross-linked nylon membrane by soaking method. The optimum immobilized conditions and the characteristics of free and immobilized papain were determined. The results showed that in pH 6.0,the carrier was soaked in 2 mg/mL papain solution at 4 ℃ for 5 h,the activity of immobilized papain 303.4U/g.The pH and temperature of optimum reaction of immobilized papain were 6.0-7.0 and 65 ℃.The pH and thermal stabilities of immobilized papain were higher than those of free papain.
