Determination of adenosine and cordycepin in culture medium of Cordyceps militaris by HPLC

Abstract

OBJECTIVE To establish an HPLC method for determining the contents of adenosine and cordycepin in Cordyceps militaris.METHODS The column was Waters Nova-Pak C_(18)(3.9 mm×300 mm,4 μm),and mobile phase was water-methanol-acetic acid(185:14:1) with flow rate of 1.0 ml·min~(-1) and detection wavelength of 260 nm.RESULTS The linear range was(2.5-)50.0 μg ·ml~(-1)(r=0.9999) for adenosine and 6.5-130.0 μg ·ml~(-1)(r=0.9999) for cordyecepin,respectively.The average recovery of adenosine was 104.5% with RSD of 1.316%;and that of cordycepin was 106.1% with RSD of 1.46%.CONCLUSIONThis method is rapid and accurate.It used to determine the adenosine and cordycepin in culture medium with satisfactory results.
