Abstract
OBJECTIVE:To observe the effect of SH2-B on motility and invasion of colon cancer cells,and detect the metastic mechanism of colon cancer.METHODS:Colon cancer cell line with a low-expressed SH2-B was selected using immunofluorescence assay.pcDNA3.1-SH2-B was transfected into HT-29 cells using Lipofectamine2000TM.Wound healing assay was used to measure HT-29 cell motility,and trans-well assay was used to test the invasion of HT-29 cell after SH2-B transfection.RESULTS:Immunofluorescence assay results showed that HT-29 cells expressed SH2-B at a relatively low Level.After HT-29 cells being transfected with pcDNA3,SH2-B expression significantly increased.Wound healing assay data showed that the moving cell numbers of the transfected,the vector and the blank groups were respectively 867±187,349±121 and 279±158(10 random fields).Compared with the controls,the moving cells increased after transfection with SH2-B(P0.05).Trans-well assay data showed that invasion cell numbers of the transfected,the vector and the blank groups were respectively 278±107,129±88 and 112±81,and invasion cells was more in the transfected group than that in the blank and the vector groups(P0.05)respectively.CONCLUSION: SH2-B may be involveed in colonal cancer metastasis through ehancing its motility and invasion.
