A preliminary study of transforming growth factor-β1-mRNA and connective tissue growth factor-mRNA on pulmonary interstitial fibrosis in rats

Abstract

Objective To explore the effect of transforming growth factor-β1-mRNA and connective tissue growth factor-mRNA on pulmonary Interstitial fibrosis in rats. Methods 48 female wistar rats were assigned randomly into 2 groups:model group and normal control group. There were 24 rats in each group. Models were founded by Bleomycin A2. The rats in the model group were filled with bleomycin A2 (5 mg/kg) once in the trachea. The normal control group were filled with equal saline in the trachea. The rats were sacrificed under drugged state at 3,7,14,28 days of feeding,then the pulmonary coefficients were observed in each group. Hematoxylin-eosin staining and van gieson chan biter acid fuchinacid staining were done and the expression of the transforming growth factor β1-mRNA and connective tissue growth factor-mRNA were detected by the nuclic acid by hybridization in situ technique. Results The expression of TGFβ1-mRNA and CTGF-mRNA in lung tissue increased as fibrosis developed. Conclusion The expression of TGFβ1-mRNA,CTGF-mRNA in lung tissue plays important role in our flowing study.
