Abstract
Mice were inoculated by gE/gI gene deleted PR inactivated propolis and oil vaccines through leg muscle injection. The muscle tissues on the injection site were collected in the day 2,7,14 and 35 PI. Paraffin tissue section were made by the normal method. The result were:The propolis vaccine was absorbed quickly after injection;There were no abnormal changes compared with the control group; Micoscopically, 2 days PI, light changes in the muscle were observed;7 days PI,there was a kind of macrophage with propolis granules in the cell plasma; More and more such macrophages were observed 14 days PI. Till 35 days PI a mass of such macrophages was formed between the muscle fibers.While the oil vaccine can not be absorbed and stay in the muscle in the injection site as long as the whole experimental period(35 d).Microscopically, fibers rupture,edema or necrosis were very common 2 days PI;there were oil vaccine residues and neutrophil cells between the muscle; Inflammation became even more severe 14 days PI;Granuloma containing oil vaccine was formed at the end of the experiment. The result demonstrated that not only the propolis vaccine has far less effect on the injection tissue than that of oil vaccine, it can also attract a handsome of phagocytes to the injection site, thus raise the efficiency of antigen presenting.Meanwhile,the propolis vaccine was engulfed and stayed long in the cell plasma of phagocytes to exert a role of ′antigen storage′,which could induce the host immune response in a more effective and persistent manner.crosis were very comm
