Abstract
Objective To observe the effect and mechanism of probucol in non-alcoholic fatty liver disease(NAFLD). Methods Sixty-four patients with non-alcoholic fatty liver disease were randomized into experimental group(n=33) and control group(n=31).The patients in experimental group were treated with probucol and diet controlling,while the patients in control group received dietary intervention and multivitamins.The period of treatment was 3 months.FPG,ALT,INS,HOMA-IRI,TNF-α,IL-8,and IL-18 of the patients in two groups were examined before and after the treatment.Results After the treatment,TG,TC,LDL-C,and ALT of the two group were decreased significantly than those before the treatment,and the difference was statistically significant(P0.05~0.001).TG,TC,LDL-C,and ALT of experimental group were declined significantly than those of control group(P0.05~0.01).The HOMA insulin resistance indexes of the two groups were both decreased after three months treatment,there were statistically significant differences between before and after the treatment(P0.05~0.001);the effect was more evident in experimental group than in control group(P0.05).There were statistically significant differences in the levels of TNF-α,IL-8 and IL-18 of experimental group between before and after the treatment(P0.01~0.001).The levels of TNF-α,IL-8 and IL-18 in experimental group and control group were decreased significantly after the treatment(P0.01~0.001),there were statistically significant differences between experimental group and control group(P0.01). Conclusions Probucol can effectively decrease TG,TC,and down-regulate the expression of TNF-α,IL-8,and IL-18 in patient with NAFLD.It is helpful to promote the liver function recovery by modulating liver fat metabolism and inhibiting liver histological activity.
