Abstract
Objective To establish a high-throughput method of DPPH free radical scavenging for evaluating the anti-oxidant activity of the extracts from Curcuma longa.Methods Taking IC50 as the evaluating criterion of DPPH free radical scavenging capability by determining the scavenging rate curves of ascorbic acid, the DPPH free radical scavenging capability of two extracts from C.longa was determined.Results At the wavelength of 515 nm, DPPH free radical scavenging rate of ascorbic acid and concentration could fit a good linearity (0.062 5—0.75 mg/mL),and the results revealed that the method is convenient and reliable.Under this condition,the DPPH free radical scavenging capability of alcohol extracts (IC50 7.78 mg/mL) was significantly stronger than that of water extracts (IC50 14.84 mg/mL) from C.longa.Conclusion The convenient and reliable method could be used to evaluate the anti-oxidant activity of other Chinese materia medica,as well as provide an approach and reference for high-throughput activity screening.
