Effect of JNK on the cerebral vasospasm after subarachnoid hemorrhage in rabbit

Abstract

Objective To observe the expression of c-jun N-terminal kinase(JNK) in the basiliar arteries after subarachnoid hemorrhage(SAH) in rabbits and discuss the relations between the expression of JNK and cerebral vasospasm(CVS).Methods 72 New Zealand white rabbits were randomized divided into control and SAH groups(including 1,3,5,7 and 10 d,n=12).A steady model of cerebral vasospasm after SAH in rabbits was produced by two-time blood injection of autologous arterial blood into the cisterna magna.HE staining and Western blot were respectively used to observe the morphological changes and the expressions of JNK in the basilar artery.The diameter of each basilar arterial lumen was measured to evaluate the degree of cerebral vasospasm.Results The basilar artery in SAH groups existed corresponding pathological changes,this model showed biphasic patterns to the luminal narrowing:acute constriction and delayed vasospasm phase.The expressions of JNK in SAH groups were increased compared to control group,and achieved peak at 7 days after SAH(P0.01),then decreased 10 days after SAH,but it was still significantly higher than that of control group(P0.01).Conclusions The expression of JNK is obvious increased and shows the regular change in time course,which is in proportion to the extent of delayed cerebral vasospasm,and it is important significance in the course of occurrence and development after CVS.
