Effect of high-fat diet on the expression of nuclear factor kappa-B in kidney of type 2 diabetic rats

Abstract
Objective To investigate the effect of hyperlipidemia on the expression of nuclear factor kappa-B(NF-κB) and intercellular adhesion molecule-1 (ICAM-1) in kidney of type 2 diabelic rats. Methods Male Wistar rats were divided into 3 groups: normal control (group A, n= 10), diabelic rats with high-fat diet (group B, n = 10) and diabetic rats with high-fat diet plus fenofibrate (group C,n=10). At the end of the 22nd week, kidneys were taken out from all rats for the measurement of expression of NF-κB P65 protein by Western blotting and expression of ICAM-1 by immunohistochemistry staining. Results The expression of NF-κB p65 protein and ICAM-1 in group C and B was significantly higher than that in group A (P0.01), and the treatment with fenofibrate significantly decreased the enhanced expression ol these two factors (C vs B, P0.01), although the expression of NF-κB P65 and ICAM-1 was still higher in group C than that in group A (P 0.01). Conclusions High-fat diet up-regulates the expression of NF-κB and ICAM-1 in kidney of diabetic rat. Fenofibrate can inhibit, although pardy, these increasing expression, which will be helpful to prevent the development of diabetic nephropathy.
