The effect of TRAIL combined with mitomycin C on apoptosis of human prostatic cancer cell DU145

Abstract

Objective To investigate the effects of TNF-related apoptosis-inducing ligand(TRAIL) and mitomycin C(MMC) on the apoptosis of human prostatic cancer cell DU145. Methods Hormone-independent prostatic cancer cell line DU145 was selected in the study.100μl of DU145 cell suspension was added to each well of 96-well plate and cultured for 24 hours.Then the cells were treated with different doses of TRAIL,MMC,or TRAIL+MMC respectively for 12 hours.Death Rates of the cells were detected by MTT test. Results The death rates of groups treated with 10ng/ml TRAIL and 25μg/ml MMC were not significantly different from that of the negative control group(P0.05).The death rates of groups treated with 50~500ng/ml TRAIL and 50~200μg/ml MMC decreased significantly when compared with the control groups(P0.05).The Rates of groups treated with combination of 50~500ng/ml TRAIL and 50~200μg/ml MMC were significantly higher than those treated with the same doses of TRAIL and MMC lonely.Although the rates did not change significantly in the groups treated with 10ng/ml TRAIL(P0.05),it could significantly increased the death rates of the groups treated with 50~200μg/ml MMC. Conclusion TRAIL can enhance the killing effect of MMC on prostatic cancer cell DU145.
